5-Fluorouracil Ameliorates the Hematotoxicity Induced by Cyclophosphamide in Male Albino Rats

Background: Cyclophosphamide (CPA) is a drug with wide spectrum of clinical uses. Its effectiveness in the treatment cancer (acute and chronic leukemias, lymphoma, multiple myeloma) non-malignant diseases such as rheumatoid arthritis vasculitis has been well established. Objectives: The present investigation aimed to study effect sub-lethal dose cyclophosphamide, 5-FU combination 5-FU, CPA on haematological parameters albino rats. Materials Methods: Twenty-eight male adults were grouped randomly into four groups (n=5 each group). Group I (control): Rats injected saline intraperitoneally at 1.0 ml/kg b.w. for 14 days. II cyclophosphamide (CPA): 10 mg/kg day by through i.p. rats III Fluorouracil (5-FU): 5-Fluorouracil was given IV (CPA+5-FU): followed per (day day) At end experimental period, anesthetized using light ether. Blood samples taken hematological evaluation. Results: White blood cells, hemoglobin content red cell counts significantly decline treated individual comparison control group, while Combination antagonize changes produced counts. Intraperitoneal caused significant reduction hematocrit platelet. reductions these measured also slightly ameliorated when animals 5-FU. individually reduced lymphocytes, neutrophils, eosinophils, monocytes; antagonized compared group. Conclusion: It could be concluded that mammals chemotherapy associated production free radicals lead hazardous alterations parameters. However, produce amelioration most cases changes. Future work should consider combined regimens, two or more mechanisms action chemotherapeutic drugs powerful than one mechanism. Using 5-fluorouracil may reduce cyclophosphamide’s side effects individually.
